DSIP (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) is a naturally occurring nonapeptide first isolated in 1974 from rabbit cerebral venous blood during induced sleep by the Schoenenberger-Monnier group. Found in the hypothalamus, limbic system, pituitary, and peripheral tissues. Research activity was highest in the 1970s–1990s; the compound is investigational with a mixed replication record.

Sleep Regulation

DSIP is studied for promotion of slow-wave (delta) sleep — the deepest sleep stage, during which GH secretion peaks and cellular repair processes are most active. Unlike melatonin (circadian timing) or sedatives (pharmacological suppression), DSIP's proposed mechanism is normalization of sleep architecture through GABAergic, opioid receptor, and HPA axis pathways.

HPA Axis Modulation — Human Data

Graf MV et al. (1984, Neuroendocrinology) documented significant attenuation of ACTH and cortisol responses to standardized stress protocols following DSIP pretreatment in human subjects. A separate study (ScienceDirect, 2000) found no significant effect on CRH/meal-induced ACTH and cortisol — indicating this mechanism has not been uniformly replicated across paradigms.
