KPV is a tripeptide (Lys-Pro-Val) corresponding to residues 11–13 of alpha-melanocyte-stimulating hormone (α-MSH). The three C-terminal amino acids retain much of α-MSH's anti-inflammatory activity while losing its pigmentary effects. KPV's mechanism is intracellular: it enters cells via the PepT1 nutrient transporter and directly inhibits NF-κB activation in the nucleus — not via melanocortin receptors.

Mechanism: PepT1-Mediated NF-κB Inhibition

The mechanistic understanding summarized here is established by the Dalmasso G et al. work cited below: KPV is transported into intestinal and epithelial cells via PepT1, and once intracellular it inhibits NF-κB — the master regulator of TNF-α, IL-1β, and IL-6 synthesis. Dalmasso G et al. (Gastroenterology. 2008;134(1):166–178) demonstrated that KPV retains its anti-inflammatory activity in MC1R-knockout mice, confirming a PepT1-mediated intracellular mechanism rather than signaling through melanocortin receptors (MCR).

Inflammatory Bowel Disease Models

Kannengiesser K et al. (Inflamm Bowel Dis. 2008;14(3):324–331) demonstrated significant anti-inflammatory effects in DSS-induced colitis and CD45RBhi transfer colitis. In the DSS model, KPV-treated animals showed earlier recovery, greater body weight regain, and reduced inflammatory infiltrates on histology. In the most severe protocol, KPV rescued all treated animals from death versus significant mortality in untreated controls.
