SNAP-8 (INCI: Acetyl Octapeptide-3; sequence: Ac-Glu-Glu-Met-Gln-Arg-Arg-Ala-Asp-NH₂) is a synthetic 8-amino-acid cosmeceutical peptide developed by Lipotec (now Lubrizol Life Science) as an extension of Argireline (Acetyl Hexapeptide-3). It is classified as a cosmetic ingredient, not a pharmaceutical, and is permitted in over-the-counter formulations under cosmetic regulations in the EU and US.
SNARE Mechanism
Muscle contraction at the neuromuscular junction requires assembly of the SNARE complex: Synaptobrevin, Syntaxin, and SNAP-25 mediate acetylcholine vesicle fusion with the neuronal membrane. SNAP-8 mimics the N-terminal end of SNAP-25 and competitively inhibits its binding in the complex, reducing acetylcholine release and attenuating facial muscle contraction signals that produce expression lines.
This is mechanistically analogous to botulinum toxin (Botox), which enzymatically cleaves SNAP-25 causing prolonged muscle paralysis. SNAP-8 works through competitive inhibition — a reversible, gentler attenuation requiring continuous daily application. The two are not equivalent in potency or mechanism. "Botox-like" is directionally accurate but misleading as to magnitude.
Clinical Evidence
Manufacturer-reported data from Lipotec (the ingredient developer, now Lubrizol Life Science), based on image-analysis of in-vivo panels, claimed wrinkle depth reductions of up to 63% over 28 days at high-concentration formulations — these are supplier-generated, not independent peer-reviewed clinical results. A separate paper in the Journal of Analytical Science and Technology (Jung JH et al., 2020) is an analytical chemistry study describing a detection/quantification method for octapeptide-3, not a clinical efficacy comparison of SNAP-8 versus Argireline; it should not be cited as evidence of superior wrinkle-reduction efficacy. Independent reviewers consistently estimate real-world effects at typical cosmetic concentrations (2–10%) in the range of 10–30% — meaningful but substantially less than the manufacturer-reported maximum values.
