Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is a synthetic heptapeptide developed at the Institute of Molecular Genetics of the Russian Academy of Sciences as a structural analog of tuftsin — a naturally occurring immunomodulatory tetrapeptide — with a C-terminal Pro-Gly-Pro extension for metabolic stability. It has been a registered pharmaceutical in Russia for anxiety disorders since the 1990s.
Primary Mechanisms
GABAergic modulation. Selank is understood to modulate GABAergic signaling, through a mechanism distinct from classical benzodiazepines, though the precise binding site has not been definitively characterized. Anxiolytic effects in animal models occur without the classical dependence pharmacology of benzodiazepines.
Serotonin metabolism. Selank is understood to influence serotonergic tone in anxiety-regulating brain regions, though the precise molecular targets (tryptophan hydroxylase, MAO-A) have not been definitively characterized in primary literature.
BDNF upregulation. Immunomodulation. As a tuftsin analog, modulates IL-6 expression and T helper cell cytokine balance — positioning Selank in psychoneuroimmunology research.
Clinical Research
Russian clinical trials evaluated Selank in generalized anxiety disorder with results supporting anxiolytic efficacy (Zozulia AA et al., Bull Exp Biol Med. 2001). Independent replication under Western controlled trial standards remains limited.
