Semax (Met-Glu-His-Phe-Pro-Gly-Pro) is a synthetic heptapeptide derived from the ACTH(4-10) sequence with a C-terminal Pro-Gly-Pro stabilizing extension, developed at the Institute of Molecular Genetics in collaboration with Moscow State University. It is a registered pharmaceutical in Russia, Belarus, and Ukraine for stroke recovery, cognitive decline, and optic nerve disease.
BDNF Upregulation
BDNF (brain-derived neurotrophic factor) is the primary mechanism of research interest. Dolotov OV et al. (Brain Res. 2006, PMID: 16996037) demonstrated Semax regulates BDNF and TrkB receptor expression in the rat hippocampus — suggesting neuroplasticity effects extending beyond acute stimulation. Intranasal administration produces CNS uptake via the olfactory pathway; animal model studies show BDNF elevation lasting up to 24 hours after a single dose.
Neuroprotection — Stroke Research
Russian clinical data document improved functional outcomes and biomarker changes consistent with enhanced neuronal survival in ischemic stroke patients treated with Semax nasal spray. Studies report upregulation of superoxide dismutase (SOD) and catalase — antioxidant enzymes protecting neurons from oxidative damage.
